Dennis Doyle (Simon Pegg) is about to marry Libby (Thandie Newton), his pregnant fiancée. However, he gets cold feet and runs away during the preparation for the wedding. Five years later, his life has gone in circles ever since. When Dennis discovers that Libby has started seeing high-flying go-getter Whit (Hank Azaria), with their son, Jake, he realizes it's now or never. He finds out that Whit is running the Nike River Marathon in London, and to prove himself to his doubting friends and, most importantly, Libby and his son, he decides to run the race himself. He receives motivation from his two "coaches", Gordon (Dylan Moran), Libby's cousin and Dennis' best friend who has made a hefty bet on Dennis succeeding, and Mr. Ghoshdastidar (Harish Patel) his landlord, who uses unorthodox methods of training, such as a spatula to spank him.
Days before the race, Whit proposes to Libby at her birthday party. Libby accepts and this puts Dennis in a state of depression. Meanwhile, Jake, who has formed a crush on a girl in his class, runs away angry when he discovers that she prefers another boy just because he has a ponytail. Dennis, upon being informed by a frightened Libby, tracks him down and explains to him that he will find many things he does not like in his life and he should just stand up to them and face them, rather than running away, having made that mistake himself, so he decides to race.

Can Dennis win the race and win back his wife??

Rock star Aldous Snow (Russell Brand) releases an album and an accompanying single "African Child". The song and music video flops, is banned and called "the worst thing to happen to Africa since the apartheid" by NME. His girlfriend, Jackie Q (Rose Byrne), takes custody of their son, Naples, and leaves him; as a result, Aldous hits rock bottom by drinking and doing drugs excessively.
Meanwhile, in Los Angeles, Aaron Green (Jonah Hill) is a recent college graduate who works as a low-level talent scout at Pinnacle Records. He lives with his girlfriend, Daphne (Elisabeth Moss), a doctor in her internship. Pinnacle Records has lost a lot of money, so the head of the company, Sergio Roma (Sean "Diddy" Combs), asks for ideas. Green proposes to have Aldous Snow play at the Greek Theatre in Los Angeles for the tenth anniversary of a particularly famous performance there. One month later, he sends Aaron to London to retrieve Aldous. Before he leaves, Daphne informs Aaron that she received a job offer at a hospital in Seattle and that they are moving there, which leads to an argument resulting in an implied break-up.

After panning across some idling diesel locomotives under the opening credits, the film begins with scenes at two rail yards in different regions of Pennsylvania run by the fictional Allegheny and West Virginia Railroad (AWVR). In the fictional Fuller yard in northern Pennsylvania, children arrive for a school field trip on rail safety. Meanwhile, in the fictional southern Pennsylvania town of Stanton, Will Colson (Chris Pine) gets up for work, stopping to surreptitiously watch his wife put their son on the school bus. He calls her but she refuses to even answer.
Arriving at work, Colson, a conductor, gets his orders for the day and learns that he will be working with engineer Frank Barnes (Denzel Washington), with whom he has never worked before. He goes to a group of older workers, with whom he has some barbed words over their age differences, and finds Barnes among them. While he goes to punch in, the other older workers complain about being displaced by Colson, who they say got his job through family connections in the union.

This movie inspired by true event,The CSX 8888 incident, also known as the Crazy Eights incident, involved an unmanned runaway train led by CSX Transportation locomotive #8888, an SD40-2, that was pulling a freight train consisting of 47 cars, some of them loaded with hazardous cargo. The train ran uncontrolled for two hours at speeds up to 51 miles per hour (82 km/h) through the U.S. state of Ohio. It was finally brought to a stop with the help of a railroad crew in a second locomotive which caught up with the runaway and coupled to the rear car.

The film opens to a young George (Jesse James) and his parents Fred (Ray Liotta) and Ermine (Rachel Griffiths). Fred files for bankruptcy and loses everything, but tries to convey to George that money is not important.
As an adult, George (Johnny Depp) moves to Southern California with his friend "Tuna" (Ethan Suplee) and they plan to earn a living by selling marijuana with the help of George's girlfriend, Barbara Buckley (Franka Potente), an airline stewardess who introduces them to Derek Foreal (Paul Reubens), the main dealer. With Derek's help, George and Tuna make a lot of money. Kevin Dulli (Max Perlich), a college student back in Boston, visits them and tells them of the enormous demand for pot in Boston. With Barbara's help, they start bringing the drugs to Boston.
